Dengue fever is an acute infectious disease caused by the dengue virus (DV), prevalent in tropical and subtropical regions worldwide and typically transmitted by mosquitoes. Dengue virus infection can manifest as asymptomatic, non-severe, or severe infection. A typical dengue fever course is divided into three phases: acute fever, critical phase, and recovery phase.

The dengue virus belongs to the genus Flavivirus, family Flaviviridae. It is an RNA virus with a single-stranded positive-sense RNA genome. There are four serotypes (DENV-1, DENV-2, DENV-3, and DENV-4), all of which can cause dengue fever infection and outbreaks. There is cross-reactivity between the different serotypes. There is also cross-reactivity with other flaviviruses such as Zika virus, Japanese encephalitis virus, and West Nile virus.

Common laboratory tests for dengue virus infection include complete blood count, blood biochemistry tests, and pathogen and serological tests. Nucleic acid testing is an important reference for dengue fever pathogen detection. China's "Dengue Fever Diagnosis and Treatment Guidelines (2014, 2nd edition)" clearly states that dengue virus nucleic acid testing can be used for early diagnosis during the acute febrile phase. For suspected or clinically diagnosed cases, detection of viral nucleic acid in acute-phase serum can serve as a basis for confirmation. China's health industry standard "Dengue Fever Diagnosis" (WS 216—2018) also includes dengue virus RNA detection as a dengue fever pathogen detection method for early diagnosis.
